According to the Twitter rules, I am confused about the messages that are considered spam.
Does it count as spam if I send a single automated message, to a new follower asking why they followed me?
Is it okay if the sender has completely positive intentions?

I’m not Twitter but I think a welcome message is a good thing to try: Overview | Docs | Twitter Developer Platform these always get through because it’s a native twitter feature and always implicitly opt in, as these get delivered when people interact for the first time

Yes, it is not allowed. There was policy clarification about this many years ago
